چکیده
In the present study, different in situ hydrogel formulations of docetaxel (DTX) based on biocompatible polymers such as Hyaluronic Acid (HA), poloxamer-407, chitosan and gellan gum were formulated to increase its therapeutic efficacy reduce toxicity. DTX was loaded nanovesicles (20 mg/mL equivalent commercial strength) further incorporated into bases possess a dual rationale protection against burst release enhanced solubility drug. The optimized formulation (NV-TPGS-3-GG-4) showed ideal rheological behavior characteristics at 37±0.5°C with sustained more than 144 h. had instant vitro gelation (2.8±0.3 min) good injectability comparison conventional injectable having (<2 h). Additionally, developed exhibited an improved biodisponibility (25.1±0.2 h) commercially available (1.7±0.1 Solid Tumor Carcinoma model Swiss albino mice revealed that (based gum) better tumor reduction (85.7±1.2%) lower toxicity compared (77.3±1.3%). Pharmacokinetic biodistribution studies demonstrated 3 4 times higher localization drug tumors. Our findings suggested gum-based can be effective delivery system for solubility, reduced toxicity, targeting tumors therapeutics. Graphical abstract

A novel ocular delivery of brinzolamide based on gellan gum: in vitro and in vivo evaluation
Background The aim of the study was to develop a sustained ocular delivery of brinzolamide (BLZ) based on gellan gum. Methods The formulations were characterized for clarity, gelling capacity, rheological studies, pH, drug content, and in vitro drug-release behavior. In vivo rabbit eye irritation test was conducted to evaluate irritation of the BLZ gel drug-delivery system. The prepared BLZ f...
متن کاملInjectable gellan gum-based nanoparticles-loaded system for the local delivery of vancomycin in osteomyelitis treatment
Infection spreading in the skeletal system leading to osteomyelitis can be prevented by the prolonged administration of antibiotics in high doses. However systemic antibiotherapy, besides its inconvenience and often low efficacy, provokes numerous side effects. Thus, we formulated a new injectable nanoparticle-loaded system for the local delivery of vancomycin (Vanc) applied in a minimally-inva...
متن کاملFormulation and In vitro Evalution of Gellan Gum/Carbopol and Sodium Alginate based Solution to Gel Depot of Ketotifen Fumarate System
Conventional ophthalmic solutions often eliminate rapidly after administration and cannot provide and maintain an adequate concentration of the drug in the precorneal area. To solve these problems, we developed a pH in situ gelling and mucoadhesive ophthalmic drug delivery system containing ketotifen fumarate based on gellan gum. The pasture of ocular drug delivery is one of the fascinating and...
متن کاملA novel in situ gel base of deacetylase gellan gum for sustained ophthalmic drug delivery of ketotifen: in vitro and in vivo evaluation
In this study, an ion-activated ketotifen ophthalmic delivery system was developed by using a natural polysaccharide, deacetylase gellan gum. Its rheological characteristics, stability, in vitro gelation, release in vitro, and pharmacodynamic activity in vivo were investigated. The formulation had an optimum viscosity that will allow easy drop as a liquid, which then underwent a rapid sol-gel t...
متن کاملIn-Situ Gelling System based on Thiolated Gellan Gum as New Carrier for Nasal Administration of Dimenhydrinate
The purpose of the present study was to develop intranasal delivery system of dimenhydrinate using thiolated gellan gum and formulations were modulated so as to have gelation at physiological ion content after intranasal administration. Gelation was determined by physical appearance. The mucoadhesive force in terms of detachment stress, determined using sheep nasal mucosa, increased with increa...
